Carry out flow cytometry and transcriptional profiling experiments to test the impact of conditional deficiency of MHC-II in specific HAPC subsets on negative selection and Treg selection throughout the perinatal to juvenile transition (Aims 2a and 3).Carry out 2-photon imaging and in vitro selection experiments (Aim 3) to determine if the perinatal thymus does not efficiently induce negative selection and if altered APC interactions contribute.Carry out in vitro and in vivo Treg suppression assays, as well as functional assays of T cell autoreactivity.Analyze data for all above experiments, prepare presentations on data for our group, participate in all relevant group meetings/journal clubs, coordinate data analysis with other investigators on the P01 and write manuscripts describing findings for timely dissemination to the research community.
